
After many failed attempts, and fruitless ventures, into finding a suitable replacement for Kratom, I’ve found 2 that are acceptable. Both have documented, and well researched, constituents, with appreciable MOR agonism. They also have other beneficial off-targets, for the goal of an energizing + mood-boosting product. The next steps will be incremental testing to find the ideal range so I can dial it in. Fortunately there are a plethora of toxicology studies on these two plants which are conveniently quite benign. Authentic M.Hirsuta has a number of interesting alkaloids including Mitraphylline, Hirsutine, Isomitraphylline, Rhynchophylline, with trace amounts of MIT, and 7-OH-MIT.
Structure Activity Relationship (SAR) analysis of the most abundant alkaloids shows poor direct MOR interactions, however numerous indirect opioid interactions through NMDA and dopamine receptor interactions. Conversely, less abundant alkaloids have direct agonistic, or partial agonistic, interactions
This was the result of a de-fatted, enzyme assisted (cellulase & pectinas), acid-base extracted, Mitragyna hirsuta. A final step consisted of using am EtOH purification and crystallization step, resulting in a 50:1 yield.

This is the result of 100 grams of Sakae Naa powder. It took approximately 2 weeks to laboriously extract.

- The release of our premier product, SR (Sleep & Recover), has hit a few obstacle, including an unintentional, and excessive, “euphoric”-like capacity, which may lead to abuse. I know that may sound ridiculous as this is a desired effect for some, however the idea behind the product was to shift away from that kind of stimulus and deliver a smooth “mood boost“ instead. Unfortunately, if that brain region is tapped to excess, parallel networks may increase in concert, leading to craving for other compounds. This is the opposite of what I intended.
- On another note, I’ve been busy in the lab extracting alkaloids from various, sometimes exotic, plants, with pretty amazing results. My current project is trying to isolate particular alkaloids in Mitragyna Hirsuta (one of Kratoms “cousins“). My last extract was a little too GABAergic for my liking whereas the goal is an uplifting and mood boosting isolate. My experiments with Mitragyna Javanica hit similar roadblocks but I will not be deterred.
